Oratie=20 professor Vermes over cel- en weefseltechnologie

Bio-engineering van = bloedvaten

Een bloedvatprothese moet de functies = van een=20 echt bloedvat kunnen overnemen. Daarom wordt zo'n prothese zoveel = mogelijk=20 van biologische materialen gemaakt. Volgens professor Vermes = houden=20 weefseltechnologen zich teveel bezig met de ontwikkeling van = stamcellen,=20 nodig voor de opbouw van het bloedvat, en te weinig met het = vaatskelet=20 waar de cellen op groeien. Prof. dr. Istv=C3=A1n Vermes geeft = tijdens zijn rede=20 ter aanvaarding van het ambt van hoogleraar Moleculaire aspecten = van cel-=20 en weefseltechnologie aan de Universiteit Twente (UT) zijn visie = op de=20 bio-engineering van bloedvaten.

Bloedvatprothesen=20 werken het beste als de biochemische en mechanische eigenschappen = zo goed=20 mogelijk overeenkomen met de werkelijkheid en als ze uit = biologisch=20 afbreekbaar materiaal bestaan. Om dit te bereiken kweken=20 weefseltechnologen onder speciale stromingscondities natuurlijke=20 vaatwandcellen, de endotheelcellen, in een biologisch afbreekbaar = buisje=20 van collageen. Volgens Professor Istv=C3=A1n Vermes is de sleutel = van het=20 succesvol ontwikkelen van een bloedvatprothese het poreuze = vaatskelet dat=20 als raamwerk dient voor de stamcellen.

Ontwikkeling van echt=20 bloedvat

Vermes:=20 "De traditionele methode gaat uit van de ontwikkeling van een = prothese=20 gemaakt van kunststof materialen. Ik houd me bezig met de = bio-engineering=20 van een bloedvat, uitgaande van biologische materialen. Daarvoor = moeten we=20 alle natuurlijke functies van een vat nabootsen, ook die van het=20 vaatskelet met alle functionele biologische materialen zoals=20 groeifactoren. Het skelet heeft veel meer functies dan enkel en = alleen het=20 aanhechten en bijeenhouden van cellen. Het bezit informatie in de = vorm van=20 groeifactoren, cytokinen en oppervlakte-eigenschappen voor de = groei en=20 ontwikkeling van cellen. De chemie, de vorm en de wijze waarop het = beweegt=20 onder invloed van stress zijn van wezenlijk belang voor het = be=C3=AFnvloeden=20 van het gedrag van cellen. Het skelet geeft signalen af, die naar = de=20 binnenzijde van de cel worden doorgegeven via receptoren op het=20 celoppervlak.=E2=80=9D De toekomst van een stamcel, hoe deze zich = ontwikkelt of=20 juist ten gronde gaat door apoptose (celdood), is afhankelijk van = de=20 informatie die door het vaatskelet wordt afgegeven.

Opbouw van de vaatwand. Een = monolaag van=20 endotheelcellen (A) en het subendotheel (B) vormen de = binnenkant. De=20 elastische tussenlagen bestaan uit de lamina elastica = interna (C),=20 gladde spiercellen (D), een matrix, en de lamina elastica = externa=20 (E). Aan de buitenkant is er een dunne laag van fibreus = bindweefsel=20 (F) en de vasa vasorum (G), de lymfevaten en zenuwen.=20

Een=20 bloedvat is vanuit de binnenkant opgebouwd uit zes verschillende = lagen van=20 achtereenvolgens endotheelcellen, elastische lagen van onder = andere gladde=20 spiercellen met daaromheen bindweefsel met lymfevaten en zenuwen. = Vermes:=20 =E2=80=9CEndotheelcellen zijn belangrijk bij het vertalen van = veranderingen in het=20 bloed via de productie van stoffen die op hun beurt zorgen voor = het=20 evenwicht tussen bloed en de omliggende weefsels. Om zicht te = krijgen op=20 de functie van deze cellen in het bloedvat en voor de productie = van een=20 kunstvat, bestuderen wij dit proces door ons te richten op de = proliferatie=20 (celdeling, red.) en apoptose van endotheelcellen.=E2=80=9D = Vermes=E2=80=99 strategie is=20 het kweken van stamcellen die zich selectief differenti=C3=ABren = tot gladde=20 spiercellen en endotheelcellen, en het ontwikkelen van een = vaatskelet in=20 de vorm van een poreuze buis van biologisch afbreekbare en = flexibele=20 polymeren. De stamcellen worden gezaaid in het skelet in de = aanwezigheid=20 van onder meer groeifactoren.
